Should grandparents voice objection to 16-year old granddaughter dating a 21-year old man?

They can voice objection because it really shouldnt have much of a bearing if the parents are okay with it.

I think it depends on the situation. If it was a 21 yr old family friend turned romantic interest, the grandparents should not say anything.

However if it was some random 21 yr old that the 16 yr old met then absolutely the grandparents should be allowed to say something.... ONCE! Voice your concerns and then leave it up to the parents.
